การผจญภัยครั้งใหม่ของ อินเดียน่า โจนส์ (แฮริสัน ฟอร์ด) เกิดขึ้นในทะเลทรายแถบตะวันตกเฉียงใต้ของอเมริกาในปี 1957 ในช่วงที่สงครามเย็นประทุถึงขีดสุด อินเดียน่า โจนส์และสหายคู่ใจของเขา แม็ค (เรย์ วินสโตน) เกือบจะเอาชีวิตไม่รอดจากการตามล่าของสายลับโซเวียต ในที่สุดเมื่ออินดี้ได้กลับมายังมหาวิทยาลัยมาร์แชล เรื่องกลับยิ่งเลวร้ายลงเมื่อเพื่อนสนิทของเขาที่เป็ นคณบดีบอกข่าวร้ายกับเขาว่ารัฐบาลสงสัยในความประพฤติ ของเขาและทางมหาวิทยาลัยได้รับแรงกดดันจากรัฐบาลให้ไ ล่เขาออก ในระหว่างที่โจนส์เดินทางเข้าเมือง เขาได้พบกับ มัตต์ (ไชอา ลาบัฟฟ์) ผู้ซึ่งพบความแค้นพร้อมกับข้อเสนอที่อินดี้ไม่สามารถ ปฏิเสธได้ ถ้าอินดี้ได้รับข้อเสนอของมัตต์โดยที่เขาต้องเสี่ยงช ีวิตอย่างที่ไม่เคยพบเจอมาก่อน อินดี้จะสร้างประวัติศาสตร์ในการค้นพบที่ยิ่งใหญ่ที่ สุดของมวลมนุษยชาติ นั่นคือกระโหลกแก้วแห่ง อาคาธอร์ โบราณวัตถุในตำนานที่ทุกคนหมายปองจะได้มาครอบครองเพร าะมันเป็นขุมพลังอำนาจ ความลี้ลับและความน่าเกรงขาม แต่การเดินทางของพวกเขาไม่ราบรื่นอย่างที่คิด เมื่อทั้งคู่พบว่าพวกเขาไม่ใช่คนเพียงกลุ่มเดียวที่ต ้องการกะโหลกแก้ว แต่เหล่าทหารโซเวียตก็ต้องการมันเพื่อยึดครองโลกด้วย เช่นกัน ด้วยเหตุนี้จึงทำให้อินดี้และมัตต์ต้องค้นหากะโหลกแก ้วให้พบก่อนทหารโซเวียตจะได้มันไปครอบครอง
