หลังจากที่ บริษัทเวชภัณฑ์และอาวุธชีวภาพ Umbrella Corporation ที่ตั้งอยู่ในเมือง “แร็กคูน ซิตี้” ล่มสลายลงด้วยคำสั่งจากรัฐบาลให้ยิงจรวดทำลายล้างเมื องเพื่อกำจัดเหล่าผู้ ติดเชื้อ t-Virus ให้สิ้นซากในวันที่ 1 ต.ค.ปี1998

หลังจากนั้นในอีก 7 ปีต่อมา ณ สนามบินแห่งหนึ่งในสหรัฐอเมริกา ตอนบ่ายของฤดูใบไม้ร่วงที่ดูเป็นปกติ กลับเกิดเหตุที่ไม่คาดฝันขึ้น เมื่อมีเครื่องบินพุ่งเข้ามาที่รอผู้โดยสารและมีผู้ต ิดเชื้อไวรัสมรณะคน หนึ่งปรากฏกายขึ้น ตามมาด้วยการแพร่กระจายเชื้อจนมีผู้กลายเป็นซอมบี้เต ็มสนามบิน ขณะที่ตัวเอกของเรื่องเป็นพระเอกหนุ่ม “Leon S. Kennedy” กับสาว “Claire Redfield” ทั้งคู่มีเวลาจำกัดแค่ 4 ชั่วโมงเท่านั้น ในการช่วยเหลือมวลมนุษย์และปฏิบัติภารกิจนี้ให้สำเร็ จ
