เริ่มต้นเมื่อ ฌอน แอนเดอร์สัน (จอช ฮัตเชอร์สัน) ได้รับรหัสสัญญาณขอความช่วยเหลือจากเกาะลึกลับ ที่นั่นมีสิ่งมีชีวิตที่มีลักษณะประหลาดทั้ง ปลาไหลเพชรฆาต ช้างตัวจิ๋ว กองไข่รูปทรงประหลาด ผึ้งยักษ์ กิ้งก่ายักษ์ แถมด้วย ภูเขาทองคำ ภูเขาไฟมรณะ ฌอนกับพ่อเลี้ยงคนใหม่ แฮงค์ (ดเวย์น เดอะ ร็อค จอห์นสัน) จึงร่วมกันออกค้นหาเกาะลึกลับแห่งนี้ พร้อมกับนักบินเฮลิคอปเตอร์ (ลูอิส กุซแมน) และลูกสาวแสนสวย (วาเนสซ่า ฮัดเจนส์) ก่อนที่แรงสั่นสะเทือนของแผ่นดินไหวจะทำให้เกาะจมอยู ่ใต้ทะเลและฝังทรัพย์ สมบัติต่างๆ ไว้ตลอดกาล
