หนังเล่าเรื่องราวของนินจาตระกูล อิงะ และ โคงะ ในยุคโตกุกาว่า โอโบโระ (ยูกิเอะ นากามะ) หลานสาวของโอเกน หัวหน้านินจาตระกูลอิงะ เจนโนสุเกะ (โจ โอดากิริ) ลูกชายของดันโจ โคงะ หัวหน้าตระกูลโคงะผู้ทรงอำนาจ ทั้งสองตระกูลอาศัยอยู่ในหุบเขาและฝึกวิทยายุทธจนมีว ิชาเหนือมนุษย์ ทั้งสองตระกูลมีความแค้นต่อกันมายาวนานหลายศตวรรษ วันหนึ่งโชกุนเทนกาอิเห็นว่านินจาเป็นอุปสรรคต่อแผนก ารยึดอำนาจของเขาจึงคิดกำจัดนินจาให้หมดไปจากญี่ปุ่น เทนกาอิจัดการประลองยุทธระหว่างนินจาสองตระกูลนี้ขึ้ น โดยให้แต่ละตระกูลส่งตัวแทนมา 5 คนเพื่อสู้กัน แต่การประลองครั้งนี้ไม่ใช่การประมือกันฉันท์มิตร หากเป็นการประลองเพื่อล้างตระกูล และการดวลครั้งนี้ก็ไม่ใช่การต่อสู้ของคนธรรมดา แต่เป็นการพบกันของนินจาที่มีวิชาเหนือมนุษย์ ไม่ว่าจะเป็น คิซารากิ ที่สามารถเปลี่ยนหน้าได้, ยาชามารุ ที่ยืดแขนขาได้, คาเกโร่ ที่ปล่อยพิษได้ หรือ ยากุชิจิ ที่มีตาวิเศษมองเห็นทุกอย่าง
