Destroy All Humans! ผู้เล่นจะได้รับบทเป็นมนุษย์ต่างดาว Crypto-137 ที่เข้ามาบุกโลกในยุคปี 1950 เพื่อสกัด DNA ของเหล่ามนุษย์ และทำลายล้างรัฐบาลสหรัฐให้ซิ้นซาก ตัวเกมเปิดโอกาสให้ผู้เล่นได้สำรวจโลกในยุคคลาสสิคด้ วย Jetpack ติดหลังสุดเท่ ต่อสู้กับกองกำลังทหารของโลกด้วยอาวุธสุดล้ำและจานบิ นจู่โจม หรือใช้พลังจิตในการควบคุมเหล่าสิ่งมีชีวิต รวมถึงการปลอมตัวเป็นมนุษย์เพื่อทำภารกิจให้ลุล่วง
