ลึกเข้าไปในป่าของโรมาเนีย นักวิทยาศาสตร์กลุ่มหนึ่งนำโดย ดร. เคทเทอรีน [Lena Headey] บังเอิญได้พบซากปรักหักพังของอารยธรรมจากศตวรรษที่ 13 หลังจากสำรวจดูพวกเค้าค้นพบสิ่งที่น่าตื่นเต้นแห่งนี ้ถูกสร้างขึ้นเหนือ ประตูทางเข้าที่นำไปสู่ถ้ำวกวนใต้พื้นดินขนาดใหญ่ นักชีววิทยาท้องถิ่นเชื่อกันว่าถ้ำแห่งนี้คือต้นกำเน ิดของระบบนิเวศวิทยา ที่ยังไม่เคยมีใครค้นพบ ดังนั้นพวกเขาจึงจ้างกลุ่มนักสำรวจถ้ำชาวอเมริกันมาช ่วยสำรวจความลึกของถ้ำ แห่งนี้

แจ็ค [Cole Hauser] และ ไทเลอร์ [Eddie Cibrian] 2 พี่น้องตระกูลแมคอัลลิสเตอร์พร้อมด้วยลูกทีมอย่าง ท๊อป [Morris Chestnut] และ ชาร์ลี [Piper Perabo] คือนักสำรวจถ้ำมืออาชีพที่ชอบแสวงหาความตื่นเต้นพวกเ ค้าคือผู้นำทีมสำรวจ ชั้นแนวหน้าของโลกที่เดินทางมาถึงโรมาเนียพร้อมด้วยเ ครื่องมือทันสมัยที่สุด แต่เมื่อพวกเค้าเริ่มสำรวจถ้ำลึกลับแห่งนี้กลับต้องพ บกับเหตุการณ์แปลก ๆ และน่าสะพรึงกลัวหลายอย่าง เพราะถ้ำลึกลับแห่งนี้มิใช่เป็นเพียงแค่ระบบนิเวศวิท ยารูปแบบใหม่เท่านั้น แต่ยังมีสิ่งมีชีวิตสายพันธุ์ใหม่อาศัยอยู่อีกด้วยแต ่พวกมันมีจุดประสงค์ เดียวต่อกลุ่มนักสำรวจ คือการฆ่าชีวิตทุกคนที่เข้ามาในถ้ำและเปลี่ยนพวกเค้า ให้เป็นบางสิ่งที่น่า สะพรึงกลัว !!!
