เกมภาคต่อในซีรีย์แอ็คชั่นลอบสังหารมุมมองบุคคลที่หน ึ่งจากค่ายเบเธสด้า ที่คราวนี้ผู้เล่นจะได้สวมบท "เอมิลี่" ที่โตเป็นสาวเต็มวัย
สำหรับเหตุการณ์ในเกมภาคสอง จะเป็นเรื่องราวที่ดำเนินต่อจากเกมภาคแรกเป็นระยะเวล า 15 ปี ภายหลังจากที่ลอร์ดรีเจนท์ถูกกำจัดและโรคไข้ระบาดจาก หนูได้หายไปจากเมืองดันวอลล์ ทว่าผู้ปกครองคนใหม่กลับไม่ใช่รัชทายาทที่ถูกต้องตาม สิทธิ์อย่าง "เอมิลี่ คัลด์วิน" (Emily Kaldwin) แต่กลับเป็นบุคคลผู้ไม่หวังดีที่เข้ามายึดครองแย่งชิ งตำแหน่ง จึงเป็นเหตุให้องค์หญิงที่ปัจจุบันได้เติบใหญ่กลายเป ็นสาว ต้องสลัดคราบผู้ดีมาเป็นมือสังหารเจริญรอยตามนักฆ่าร ุ่นพี่ เพื่อทวงบัลลังก์ของตนกลับคืน

ตัวเกมดิสฮอเนอร์ในภาคใหม่นี้ จะเปิดโอกาสให้ผู้เล่นสามารถเลือกสวมบทเป็นได้ทั้งมื อสังหารสาวสวย "เอมิลี่ คัลด์วิน" หรือหนุ่มนักฆ่า "คอร์โว อัททาโน" (ตัวละครเอกจากภาคแรก) โดยที่ทั้งคู่จะมีสกิลทักษะ อาวุธ รวมถึงพลังเหนือธรรมชาติในการลอบสังหารที่ต่างกันไป และเหมือนภาคที่แล้วที่ผู้เล่นจะสามารถตัดสินใจเลือก วิธีเข้าจัดการกับศัตรูได้อิสระ ไม่ว่าจะลอบเร้นไม่ให้ศัตรูรู้ตัว หรือชักอาวุธปะทะกันตรงๆ ซึ่งทุกการตัดสินใจของผู้เล่นจะส่งผลกระทบต่อผลลัพธ์ ของภารกิจ หรือแม้กระทั่งฉากจบของเกม
